Chromosome loss in bi-parental progenies of tetraploid Pacific oyster Crassostrea gigas ArchiMer
Mccombie, Helen; Lapegue, Sylvie; Cornette, Florence; Ledu, Christophe; Boudry, Pierre.
Alterations of chromosome number have been observed in the somatic tissue of Crassostrea gigas diploids and artificial polyploids. Tetraploid oysters with abnormal chromosome numbers in some or all of their tissue are considered undesirable as parents either for triploids (produced via a cross with diploids and of aquacultural interest) or for tetraploid breeding. Aneuploid tetraploid oysters may confer the tendency to lose chromosomes and to revert to lower ploidy levels to their offspring. More directly, their offspring could have a lower ploidy because of potential links between somatic and gametic chromosome loss. The present study evaluated the phenomenon in six bi-parental tetraploid families bred from parents of differing somatic ploidy quality. The...

Circadian G2 arrest as related to circadian gating of cell population growth in Euglena OAK
Bolige, Aoen; Hagiwara, Shin-ya; Zhang, Yulan; Goto, Ken; 後藤, 健.
Cell population growth is gated to occur in particular circadian phases, which has been known for over four decades in various organisms including cyanobacteria and human. However, little is known as to which cell cycle phases from G(1) to M are primarily regulated by the circadian rhythm or when in a circadian cycle this primary regulation takes place. We report here that in the flagellate alga Euglena gracilis grown photoautotrophically, the circadian rhythm primarily prevented developmentally matured G(2) cells from progressing to mitosis, such that cell population growth occurred only during subjective night. In addition, we found that the circadian rhythm also arrests G(1)-to-S and S-to-G(2) transitions at particular circadian phases.

Cytogenetical and ultrastructural effects of copper on root meristem cells of Allium sativum L. Biocell
Liu,Donghua; Jiang,Wusheng; Meng,Qingmin; Zou,Jin; Gu,Jiegang; Zeng,Muai.
Different copper concentrations, as well as different exposure times, were applied to investigate both cytogenetical and ultrastructural alterations in garlic (Allium sativum L.) meristem cells. Results showed that the mitotic index decreased progressively when either copper concentration or exposure time increased. C-mitosis, anaphase bridges, chromosome stickiness and broken nuclei were observed in the copper treated root tip cells. Some particulates containing the argyrophilic NOR-associated proteins were distributed in the nucleus of the root-tip cells and the amount of this particulate material progressively increased with increasing exposure time. Finally, the nucleolar material was extruded from the nucleus into the cytoplasm. Also, increased...

Influence of 2-methoxyestradiol on MCF-7 cells: An improved differential interference contrasting technique and Bcl-2 and Bax protein expression levels Biocell
Joubert,Annie; Marais,Sumari; Maritz,Christine.
Proteins of the B-cell lymphoma 2 family are crucial for the regulation of apoptosis. B-cell lymphoma 2-associated X is a pro-apoptotic protein, while B-cell lymphoma 2 protein opposes apoptosis. The influence of 1 μM 2-methoxyestradiol was investigated on the expression levels of these two proteins in MCF-7 cells. 2-Methoxyestradiol exposure did not influence B-cell lymphoma 2 protein expression levels after 24 h of exposure. In contrast, B-cell lymphoma 2-associated X protein levels were significantly reduced. An improved differential interference contrasting technique revealed compromised cell density and the presence of a mitotic block in exposed cells. The study proposes that the influence of 2-methoxyestradiol on the expression of these...

KARYOTYPE ANALYSIS IN GRAPEVINES Rev. Bras. Frutic.
PIEROZZI,NEIVA IZABEL; MOURA,MARA FERNANDES.
ABSTRACT Seven species of Vitis L., V. champinii Planchon, V. cinerea (Engelm in Gray) Engelm, V. girdiana Munson, V. labrusca L., V. rotundifolia Michaux, V. rupestres Scheels and V. vinifera L. were studied with the purpose of complementing the karyomorphometric information for further comparative analyses. Based on ideograms and on chromosome measures obtained it was possible to see several differences among the species, which were enough to distinguish at least V. champinii and V. girdiana from the others as well as V. labrusca for the lowest measures and V. rotundifolia for the highest mean value of arm ratio. It seems that during the species diversification process the most crucial differences among them did not involve drastic changes in chromosome...

The effect of magnesium on mitotic spindle formation in Schizosaccharomyces pombe Genet. Mol. Biol.
Uz,Gulsen; Sarikaya,Aysegul Topal.
Abstract Magnesium (Mg2+), an essential ion for cells and biological systems, is involved in a variety of cellular processes, including the formation and breakdown of microtubules. The results of a previous investigation suggested that as cells grow the intracellular Mg2+ concentration falls, thereby stimulating formation of the mitotic spindle. In the present work, we used a Mg2+-deficient Schizosaccharomyces pombe strain GA2, in which two essential membrane Mg2+ transporter genes (homologs of ALR1 and ALR2 in Saccharomyces cerevisae) were deleted, and its parental strain Sp292, to examine the extent to which low Mg2+ concentrations can affect mitotic spindle formation. The two S. pombe strains were transformed with a plasmid carrying a GFP-α2-tubulin...
